TC Pool Manager

If you set a new env variable on the TC Pool Manager server, do you have to restart the service, for it to take effect?

System Environment variables are read during OS startup. The command line utility "SETX" can make the variable available immediately however current running tcserver's will not see it. You can restart Server Manager so that all tcservers get the value or you can restart only the warm servers (nobody connected) using the Admin Console and any new connections will see it (currently connected tcservers will not see it). Restarting the Server Manager service after running SETX or rebooting the server are the only ways I use to guarantee the setting is honored.
